Arriving at the Hotel Excelsior on Tuesday morning, Penelope, 52, looked stunning in a plunging black dress adorned with feathers on the shoulders.
She added dark shades and a pair of sky-high heels.
Javier, 57, was also dressed for the occasion and looked sharp in a camel suit and white shirt.
The couple flew to the Italian city for the Tuesday night premiere of Florian Zeller’s Bunker, marking the ninth time they have worked together on screen.
In the psychological thriller, the pair play married couple Sofia and Miguel.

They first met in 1992 on the set of the Spanish film Jamón, Jamón, when Penelope was 17 and Javier 23.
Although they played romantic interests in the film, they did not start dating until 15 years later when they were reunited on the set of the 2008 film Vicky Cristina Barcelona, for which Penelope won the Oscar for Best Supporting Actress.
In May, Javier shared that since the birth of their first child, he and Penelope have made sure at least one of them is home all the time when they take on acting roles.
The No Country For Old Men star praised his wife, adding that he feels “blessed” to have “had the opportunity to be in life at the same time, in the same place” as her.
He said: “I have no obligations in my contract related to larger trailers or luxury suites.
‘The only thing I do make sure is that I am not away from my family more than two weeks.’

The Venice Film Festival kicked off last Wednesday with the premiere of Ink by British director Danny Boyle, a drama about the rise of media mogul Rupert Murdoch.
It closes on September 12 with the Italian costume drama Dio Ride (God Laughs), written and directed by Giovanni Veronesi.
American actress and filmmaker Maggie Gyllenhaal chairs the jury for the main competition.
She is joined by Tunisian director and screenwriter Kaouther Ben Hania, English composer Daniel Blumberg, Italian film critic Francesco Casetti, French director Xavier Giannoli, Afghan director Shahrbanoo Sadat and Hong Kong director Johnnie To.
