According to initial reports, the two occupants of the helicopter were injured. Their nationalities are not yet known.
Despite the crash taking place on a busy highway, there are no other reports of any casualties at this stage.
Traffic has been blocked in both directions so that emergency services can arrive on the scene.
The accident is linked to heavy winds that are said to have destabilized the helicopter.
Local reports said the plane was taking part in an exhibition for the helicopter industry called European Rotors at the IFEMA exhibition center in Madrid.
Aviation news website On The Wings of Aviation reported: 'The Enstrom 280FX Shark helicopter with registration F-HPUX participating in the European Rotors exhibition held in Ifema crashed on the M40 in Madrid shortly after take-off.
“At this time it is unknown whether there are any injuries or what the causes of the accident are.”
