A young child has been killed and another seriously injured in a head-on collision in Melbourne's far south-east.

The vehicles crashed on the Maroondah Highway just after 4pm on Friday, police said.

A primary school-age child died at the scene, while another child from the same vehicle was taken to hospital with serious injuries.

Their driver was slightly injured and taken to hospital.

The driver and sole occupant of the other car were taken to hospital with minor injuries.

The Maroondah Highway between Melba Highway and St Huberts Road remains closed while major crash detectives investigate the fatal collision and process the scene.

It comes after a 12-year-old girl was killed in a separate head-on crash on the Hamilton Highway in Stonehaven, about 10km west of Geelong, on Wednesday.

Police allege the disqualified driver behind the wheel of the other car, a 28-year-old man from North Shore, was speeding and overtaking other motorists in the lead-up to the crash.

He was released from hospital on Friday and will be questioned by detectives, with charges yet to be filed.
